Various proteolytic enzymes are used to detach cells from the adherent substrate by breaking down vessel-adhering proteins to allow for easy resuspension during cell harvesting. Among the proteolytic enzymes, Trypsin, a member of serine protease family that cleaves at the C-terminal side of lysine and arginine, is most frequently used. However, long term incubation with high trypsin concentration damage cells by striping cell surface proteins and kill the cells. This sticker shows how researches feel a few minutes after adding trypsin.
